After graduating from Emory University, Dumas Malone attended and taught at Yale University. Following service in World War I, he was appointed professor at the University of Virginia in 1923. He was editor, then editor-in-chief, of "The Dictionary of American Biography" and from 1936 to 1942 was editor-in-chief of the Harvard University Press.

From 1945 to 1959, Malone was a professor of history at Columbia University. He returned to Charlottesville as Thomas Jefferson Foundation professor emeritus and biographer-in-residence in 1962. He completed "Jefferson and His Time" in 1981. He was given the Pulitzer Prize in 1975 for his "magnum opus" and in 1983 was awarded the Presidential Medal of Freedom.
